NM_000251.3(MSH2):c.294_295del (p.Tyr98_Arg99delinsTer) was classified as Pathogenic for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023. This variant lies in the MSH2 gene (transcript NM_000251.3) at coding-DNA position 294 through coding-DNA position 295, deleting 2 bases. Submitter rationale: The c.294_295delTA pathogenic mutation, located in coding exon 2 of the MSH2 gene, results from a deletion of two nucleotides at nucleotide positions 294 to 295, causing a translational frameshift with a predicted alternate stop codon (p.Y98*). This mutation was detected in 1/537 French families tested for Lynch syndrome (Bonadona V et al. JAMA, 2011 Jun;305:2304-10). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
