This variant lies in the MLH1 gene (transcript NM_000249.4) at the canonical splice donor site of the intron immediately after coding-DNA position 677, where T is replaced by C; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: For these reasons, this variant has been classified as Pathogenic. Studies have shown that disruption of this splice site results in skipping of exon 8 and introduces a premature termination codon (Invitae). The resulting mRNA is expected to undergo nonsense-mediated decay. ClinVar contains an entry for this variant (Variation ID: 525602). Disruption of this splice site has been observed in individuals with MLH1-related conditions (PMID: 12624141, 15342696, 17312306, 27601186; Invitae). This variant is not present in population databases (gnomAD no frequency). This sequence change affects a donor splice site in intron 8 of the MLH1 gene. RNA analysis indicates that disruption of this splice site induces altered splicing and may result in an absent or disrupted protein product.
