Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_001364171.2(ODAD1):c.1192G>A (p.Glu398Lys), citing Ambry Variant Classification Scheme 2023: The p.E361K variant (also known as c.1081G>A), located in coding exon 9 of the CCDC114 gene, results from a G to A substitution at nucleotide position 1081. The glutamic acid at codon 361 is replaced by lysine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
