Pathogenic for Primary ciliary dyskinesia — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_012144.4(DNAI1):c.625C>T (p.Arg209Ter), citing Invitae Variant Classification Sherloc (09022015): This variant has not been reported in the literature in individuals affected with DNAI1-related conditions. ClinVar contains an entry for this variant (Variation ID: 525320). For these reasons, this variant has been classified as Pathogenic. This sequence change creates a premature translational stop signal (p.Arg209*) in the DNAI1 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in DNAI1 are known to be pathogenic (PMID: 16858015, 29363216). This variant is present in population databases (rs769224534, gnomAD 0.02%).
