Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_178452.6(DNAAF1):c.223G>A (p.Ala75Thr), citing Ambry Variant Classification Scheme 2023: The p.A75T variant (also known as c.223G>A), located in coding exon 2 of the DNAAF1 gene, results from a G to A substitution at nucleotide position 223. The alanine at codon 75 is replaced by threonine, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
