Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_001364171.2(ODAD1):c.716AGG[1] (p.Glu240del), citing Ambry Variant Classification Scheme 2023: The c.608_610delAGG variant (also known as p.E203del), located in coding exon 6 of the CCDC114 gene, results from an in-frame AGG deletion at nucleotide positions 608 to 610. This results in the in-frame deletion of a glutamic acid at codon 203. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis (Choi Y et al. PLoS ONE. 2012; 7(10):e46688). Based on the available evidence, the clinical significance of this variant remains unclear.
