Uncertain significance for Hypertrophic cardiomyopathy — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000363.5(TNNI3):c.323A>T (p.Asp108Val), citing Invitae Variant Classification Sherloc (09022015): This variant has not been reported in the literature in individuals with TNNI3-related disease. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C15"). This variant is not present in population databases (ExAC no frequency). This sequence change replaces aspartic acid with valine at codon 108 of the TNNI3 protein (p.Asp108Val). The aspartic acid residue is highly conserved and there is a large physicochemical difference between aspartic acid and valine.
