NM_000051.4(ATM):c.737A>G (p.Asn246Ser) was classified as Uncertain significance for Familial cancer of breast by St. Jude Molecular Pathology, St. Jude Children's Research Hospital, citing St. Jude Assertion Criteria 2020. This variant lies in the ATM gene (transcript NM_000051.4) at coding-DNA position 737, where A is replaced by G; at the protein level this means replaces asparagine at residue 246 with serine — a missense variant. Submitter rationale: The ATM c.737A>G p.(Asn246Ser) missense change has a maximum subpopulation frequency of 0.003% in gnomAD v2.1.1 (https://gnomad.broadinstitute.org/). The in silico tool REVEL predic ts a benign effect on protein function, but to our knowledge this prediction has not been confirmed by functional studies. This variant has been reported in individuals with breast cancer as well (PMID: 33471991, 30613976, 33785725) as control individual s (PMID: 30303537, 30287823). To our knowledge, this variant has not been reported in individuals with ataxia telangiectasia. In summary, the evidence currently available is insufficient to determine the clinical significance of this variant. It has ther efore been classified as of uncertain significance.
