Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000051.4(ATM):c.2765A>T (p.Asp922Val), citing Ambry Variant Classification Scheme 2023: The p.D922V variant (also known as c.2765A>T), located in coding exon 17 of the ATM gene, results from an A to T substitution at nucleotide position 2765. The aspartic acid at codon 922 is replaced by valine, an amino acid with highly dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
