Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000051.4(ATM):c.871C>G (p.His291Asp), citing Ambry Variant Classification Scheme 2023: The p.H291D variant (also known as c.871C>G), located in coding exon 6 of the ATM gene, results from a C to G substitution at nucleotide position 871. The histidine at codon 291 is replaced by aspartic acid, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear.
