Likely pathogenic — the classification assigned by GeneDx to NM_002495.4(NDUFS4):c.377_384del (p.Leu126fs), citing GeneDx Variant Classification (06012015): The c.377_384delTAACCTTC variant in the NDUFS4 gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. The c.377_384delTAACCTTC variant causes a frameshift starting with codon Leucine 126, changes this amino acid to a Glutamine residue, and creates a premature Stop codon at position 3 of the new reading frame, denoted p.Leu126GlnfsX3. This variant is predicted to cause loss of normal protein function through protein truncation, as the last 50 amino acids are lost and replaced with 2 incorrect amino acids. This variant is not observed in large population cohorts (Lek et al., 2016). We interpret c.377_384delTAACCTTC as a likely pathogenic variant.
