This variant lies in the GATA6 gene (transcript NM_005257.6) at coding-DNA position 925, deleting one base; at the protein level this means shifts the reading frame starting at glutamine residue 309, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: The c.925delC variant in the GATA6 gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. The c.925delC variant causes a frameshift starting with codon Glutamine 309, changes this amino acid to a Serine residue, and creates a premature Stop codon at position 11 of the new reading frame, denoted p.Gln309SerfsX11. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The c.925delC variant is not observed in large population cohorts (Lek et al., 2016). We interpret c.925delC as a pathogenic variant.
