This variant lies in the XPA gene (transcript NM_000380.4) at coding-DNA position 327 through coding-DNA position 340, duplicating 14 bases; at the protein level this means shifts the reading frame starting at aspartic acid residue 114, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: The c.327_340dup14 variant in the XPA gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. The c.327_340dup14 variant causes a frameshift starting with codon Aspartic acid 114, changes this amino acid to a Glycine residue, and creates a premature Stop codon at position 10 of the new reading frame, denoted p.Asp114GlyfsX10. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The c.327_340dup14 variant is not observed in large population cohorts (Lek et al., 2016). We interpret c.327_340dup14 as a pathogenic variant.
