NM_007315.4(STAT1):c.419_422del (p.Lys140fs) was classified as Likely pathogenic by GeneDx, citing GeneDx Variant Classification (06012015): The c.419_422delAAGA variant in the STAT1 gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. The c.419_422delAAGA variant causes a frameshift starting with codon Lysine 140, changes this amino acid to a Serine residue, and creates a premature Stop codon at position 9 of the new reading frame, denoted p.Lys140SerfsX9. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The c.419_422delAAGA variant is not observed at a significant frequency in large population cohorts (Lek et al., 2016). We interpret c.419_422delAAGA as a likely pathogenic variant.
