NM_000059.4(BRCA2):c.7696G>T (p.Asp2566Tyr) was classified as Uncertain significance by Women's Health and Genetics/Laboratory Corporation of America, LabCorp, citing LabCorp Variant Classification Summary - May 2015: Variant summary: BRCA2 c.7696G>T (p.Asp2566Tyr) results in a non-conservative amino acid change located in the Breast cancer type 2 susceptibility protein, helical domain (IPR015252) of the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 251322 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.7696G>T in individuals affected with Hereditary Breast And Ovarian Cancer Syndrome and no experimental evidence demonstrating its impact on protein function have been reported. However, an algorithm for predicting the protein likelihood ratio of the BRCA2 gene suggested the variant is neutral (Karchin_2008). The following publication has been ascertained in the context of this evaluation (PMID: 19043619). Two ClinVar submitters have assessed the variant since 2014: one submitter classified the variant as likely benign, and one submitter classified the variant as uncertain significance. Based on the evidence outlined above, the variant was classified as uncertain significance.
