Pathogenic — the classification assigned by GeneDx to NM_003482.4(KMT2D):c.2141C>A (p.Ser714Ter), citing GeneDx Variant Classification (06012015). This variant lies in the KMT2D gene (transcript NM_003482.4) at coding-DNA position 2141, where C is replaced by A; at the protein level this means converts the codon for serine at residue 714 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: The S714X variant in the KMT2D gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The S714X variant is not observed in large population cohorts (Lek et al., 2016). We interpret S714X as a pathogenic variant.
