This variant lies in the BRCA2 gene (transcript NM_000059.4) at coding-DNA position 7583, where G is replaced by A; at the protein level this means replaces glycine at residue 2528 with glutamic acid — a missense variant. Submitter rationale: The BRCA2 c.7583G>A (p.Gly2528Glu) variant has been reported in the published literature to be located in a region of the BRCA2 gene that is tolerant to missense changes (PMID: 31911673 (2020)). To the best of our knowledge, this variant has not been reported in individuals with BRCA2-related disorders. Functional studies demonstrated that this variant was not damaging to BRCA2 protein function (PMID: 39779848 (2025), 39779857 (2025)). The frequency of this variant in the general population (Genome Aggregation Database, http://gnomad.broadinstitute.org) is uninformative in the assessment of its pathogenicity. Analysis of this variant using bioinformatics tools for the prediction of the effect of amino acid changes on protein structure and function yielded predictions that this variant is benign. Based on the available information, we are unable to determine the clinical significance of this variant.
