Uncertain significance — the classification assigned by Quest Diagnostics Nichols Institute San Juan Capistrano to NM_000059.4(BRCA2):c.2399G>A (p.Gly800Asp), citing Quest Diagnostics criteria: The BRCA2 c.2399G>A (p.Gly800Asp) variant has been reported to be located in a region of the BRCA2 gene that is tolerant to missense sequence changes in the published literature (PMID: 31911673 (2020)). In a large scale breast cancer association study, this variant has been observed in one breast cancer case but not in the cohort of reportedly healthy individuals (PMID: 33471991 (2021), see also LOVD (http://databases.lovd.nl/shared)). This variant has not been reported in large, multi-ethnic general populations (Genome Aggregation Database, http://gnomad.broadinstitute.org). Analysis of this variant using bioinformatics tools for the prediction of the effect of amino acid changes on protein structure and function yielded predictions that this variant is benign. Based on the available information, we are unable to determine the clinical significance of this variant.
