Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_005120.3(MED12):c.3303C>G (p.Cys1101Trp), citing Ambry Variant Classification Scheme 2023: The c.3303C>G (p.C1101W) alteration is located in coding exon 23 of the MED12 gene. This alteration results from a C to G substitution at nucleotide position 3303, causing the cysteine (C) at amino acid position 1101 to be replaced by a tryptophan (W). This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). This amino acid position is highly conserved in available vertebrate species. The in silico prediction for this alteration is inconclusive. Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.
