Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_021008.4(DEAF1):c.658G>A (p.Gly220Ser), citing LabCorp Variant Classification Summary - May 2015: Variant summary: DEAF1 c.658G>A (p.Gly220Ser) results in a non-conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change. The variant was absent in 251282 control chromosomes. c.658G>A has been observed in individuals affected with autism spectrum disorder, including one individual with Vulto van Silfout de Vries syndrome in whom it was reported as de novo (e.g. Lim_2017, Duan_2022). To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publications have been ascertained in the context of this evaluation (PMID: 28714951, 31785789, 31981491, 35231114). ClinVar contains an entry for this variant (Variation ID: 521587). Based on the evidence outlined above, the variant was classified as VUS-possibly pathogenic.
