Uncertain significance — the classification assigned by Ambry Genetics to NM_001719.3(BMP7):c.457del (p.His153fs), citing Ambry Variant Classification Scheme 2023: The c.457delC (p.H153Ifs*112) alteration, located in exon 2 (coding exon 2) of the BMP7 gene, consists of a deletion of one nucleotide at position 457, causing a translational frameshift with a predicted alternate stop codon after 112 amino acids. This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. However, loss of function of BMP7 has not been established as a mechanism of disease. Based on data from the Genome Aggregation Database (gnomAD), the BMP7 c.457delC alteration was not observed, with coverage at this position. Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.
