NM_003036.4(SKI):c.1439CGGAGG[1] (p.Ala482_Glu483del) was classified as Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The c.1445_1450delCGGAGG variant (also known as p.A482_E483del) is located in coding exon 4 of the SKI gene. This variant results from an in-frame CGGAGG deletion at nucleotide positions 1445 to 1450. This results in the in-frame deletion of two amino acids at codons 482 and 483. This amino acid region is highly conserved in available vertebrate species. In addition, the in silico prediction for this variant is inconclusive (Choi Y et al. PLoS ONE. 2012; 7(10):e46688). Based on the available evidence, the clinical significance of this variant remains unclear.
