NM_004415.4(DSP):c.8309A>G (p.Tyr2770Cys) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Y2770C variant (also known as c.8309A>G), located in coding exon 24 of the DSP gene, results from an A to G substitution at nucleotide position 8309. The tyrosine at codon 2770 is replaced by cysteine, an amino acid with highly dissimilar properties. This variant was reported in individual(s) with features consistent with arrhythmogenic right ventricular cardiomyopathy (ARVC) (Walsh R et al. Genet. Med., 2017 02;19:192-203; Gasperetti A et al. JACC Adv. 2024 Mar;3(3):100832). This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
