This variant lies in the PTPN11 gene (transcript NM_002834.5) at coding-DNA position 1027, where C is replaced by T; at the protein level this means replaces arginine at residue 343 with tryptophan — a missense variant. Submitter rationale: The p.R343W variant (also known as c.1027C>T), located in coding exon 9 of the PTPN11 gene, results from a C to T substitution at nucleotide position 1027. The arginine at codon 343 is replaced by tryptophan, an amino acid with dissimilar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6503 samples (13006 alleles) with coverage at this position. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
