Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_004612.4(TGFBR1):c.290C>A (p.Thr97Lys), citing Ambry Variant Classification Scheme 2023: The p.T97K variant (also known as c.290C>A), located in coding exon 2 of the TGFBR1 gene, results from a C to A substitution at nucleotide position 290. The threonine at codon 97 is replaced by lysine, an amino acid with similar properties, and is located in the extracellular domain. This amino acid position is not well conserved in available vertebrate species, and lysine is the reference amino acid in other vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
