Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_003239.5(TGFB3):c.1055G>A (p.Arg352His), citing Ambry Variant Classification Scheme 2023. This variant lies in the TGFB3 gene (transcript NM_003239.5) at coding-DNA position 1055, where G is replaced by A; at the protein level this means replaces arginine at residue 352 with histidine — a missense variant. Submitter rationale: The p.R352H variant (also known as c.1055G>A), located in coding exon 6 of the TGFB3 gene, results from a G to A substitution at nucleotide position 1055. The arginine at codon 352 is replaced by histidine, an amino acid with highly similar properties. Based on data from ExAC, the A allele has an overall frequency less than 0.01% (1/106184). This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6503 samples (13006 alleles) with coverage at this position. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
