NM_003238.6(TGFB2):c.764G>A (p.Gly255Asp) was classified as Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.G255D variant (also known as c.764G>A), located in coding exon 5 of the TGFB2 gene, results from a G to A substitution at nucleotide position 764. The glycine at codon 255 is replaced by aspartic acid, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species, and aspartic acid is the reference amino acid in other vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
