Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_003238.6(TGFB2):c.410T>C (p.Met137Thr), citing Ambry Variant Classification Scheme 2023. This variant lies in the TGFB2 gene (transcript NM_003238.6) at coding-DNA position 410, where T is replaced by C; at the protein level this means replaces methionine at residue 137 with threonine — a missense variant. Submitter rationale: The p.M137T variant (also known as c.410T>C), located in coding exon 2 of the TGFB2 gene, results from a T to C substitution at nucleotide position 410. The methionine at codon 137 is replaced by threonine, an amino acid with similar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6503 samples (13006 alleles) with coverage at this position. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
