Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_003238.6(TGFB2):c.548G>A (p.Arg183His), citing Ambry Variant Classification Scheme 2023: The p.R183H variant (also known as c.548G>A), located in coding exon 3 of the TGFB2 gene, results from a G to A substitution at nucleotide position 548. The arginine at codon 183 is replaced by histidine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
