The methionine residue is highly conserved (100 vertebrates, UCSC), and is located in the MH2 domain a region, amino acids 232-425, that is defined as a mutational hotspot and critical functional domain (PMID: 30661052). There is a moderate physicochemical difference between methionine and lysine. This variant is absent from gnomAD v2.1 and v3.1. This variant has been reported in at least three probands with thoracic aortic aneurysm or dissection (PMID: 29543232; ClinVar: SCV001992489.1; Royal Melbourne Hospital). Multiple lines of computational evidence predict a deleterious effect for the missense substitution (6/6 algorithms).
