Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_030777.4(SLC2A10):c.1018G>A (p.Gly340Ser), citing Ambry Variant Classification Scheme 2023. This variant lies in the SLC2A10 gene (transcript NM_030777.4) at coding-DNA position 1018, where G is replaced by A; at the protein level this means replaces glycine at residue 340 with serine — a missense variant. Submitter rationale: The p.G340S variant (also known as c.1018G>A), located in coding exon 2 of the SLC2A10 gene, results from a G to A substitution at nucleotide position 1018. The glycine at codon 340 is replaced by serine, an amino acid with similar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6503 samples (13006 alleles) with coverage at this position. This amino acid position is not well conserved in available vertebrate species, and serine is the reference amino acid in other vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this variant remains unclear.
