NM_000302.4(PLOD1):c.214G>T (p.Ala72Ser) was classified as Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection by Ambry Genetics, citing Ambry Variant Classification Scheme 2023. This variant lies in the PLOD1 gene (transcript NM_000302.4) at coding-DNA position 214, where G is replaced by T; at the protein level this means replaces alanine at residue 72 with serine — a missense variant. Submitter rationale: The p.A72S variant (also known as c.214G>T), located in coding exon 3 of the PLOD1 gene, results from a G to T substitution at nucleotide position 214. The alanine at codon 72 is replaced by serine, an amino acid with some similar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6503 samples (13006 alleles) with coverage at this position. This amino acid position is not well conserved in available vertebrate species; however, serine is the reference amino acid in other vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significant of this variant remains unclear.
