This variant lies in the NOTCH1 gene (transcript NM_017617.5) at coding-DNA position 5914, where G is replaced by A; at the protein level this means replaces aspartic acid at residue 1972 with asparagine — a missense variant. Submitter rationale: The p.D1972N variant (also known as c.5914G>A), located in coding exon 31 of the NOTCH1 gene, results from a G to A substitution at nucleotide position 5914. The aspartic acid at codon 1972 is replaced by asparagine, an amino acid with highly similar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6475 samples (12950 alleles) with coverage at this position. This amino acid position is highly conserved in available vertebrate species.
