This variant lies in the NOTCH1 gene (transcript NM_017617.5) at coding-DNA position 59, where G is replaced by T; at the protein level this means replaces arginine at residue 20 with leucine — a missense variant. Submitter rationale: The p.R20L variant (also known as c.59G>T), located in coding exon 1 of the NOTCH1 gene, results from a G to T substitution at nucleotide position 59. The arginine at codon 20 is replaced by leucine, an amino acid with dissimilar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. This position was not covered in the ESP. This amino acid position is not well conserved on limited sequence alignment.
