Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_001110556.2(FLNA):c.1698C>G (p.Phe566Leu), citing Ambry Variant Classification Scheme 2023: The p.F566L variant (also known as c.1698C>G), located in coding exon 11 of the FLNA gene, results from a C to G substitution at nucleotide position 1698. The phenylalanine at codon 566 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
