Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_001110556.2(FLNA):c.6763G>C (p.Val2255Leu), citing Ambry Variant Classification Scheme 2023. This variant lies in the FLNA gene (transcript NM_001110556.2) at coding-DNA position 6763, where G is replaced by C; at the protein level this means replaces valine at residue 2255 with leucine — a missense variant. Submitter rationale: The p.V2247L variant (also known as c.6739G>C), located in coding exon 39 of the FLNA gene, results from a G to C substitution at nucleotide position 6739. The valine at codon 2247 is replaced by leucine, an amino acid with highly similar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6217 samples with coverage at this position. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this variant remains unclear.
