This variant lies in the FLNA gene (transcript NM_001110556.2) at coding-DNA position 5506, where G is replaced by A; at the protein level this means replaces glutamic acid at residue 1836 with lysine — a missense variant. Submitter rationale: The p.E1828K variant (also known as c.5482G>A), located in coding exon 32 of the FLNA gene, results from a G to A substitution at nucleotide position 5482. The glutamic acid at codon 1828 is replaced by lysine, an amino acid with similar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6428 samples with coverage at this position. This amino acid position is highly conserved in available vertebrate species; however, lysine is the reference amino acid in other vertebrate species.
