Submitter rationale: This missense variant replaces proline with leucine at codon 2 of the BRCA2 protein. Computational prediction suggests that this variant may not impact protein structure and function (internally defined REVEL score threshold <= 0.5, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has been reported in two individuals affected with breast cancer (PMID: 35264596, 35864222). This variant has been reported in a multifactorial analysis with co-occurrence and family history likelihood ratios for pathogenicity of 1.025 and 0.653, respectively (PMID: 31131967). This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD).
