NM_000138.5(FBN1):c.2898G>C (p.Glu966Asp) was classified as Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E966D variant (also known as c.2898G>C), located in coding exon 24 of the FBN1 gene, results from a G to C substitution at nucleotide position 2898. The glutamic acid at codon 966 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
