Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_000138.5(FBN1):c.1639A>G (p.Ile547Val), citing Ambry Variant Classification Scheme 2023: The p.I547V variant (also known as c.1639A>G), located in coding exon 13 of the FBN1 gene, results from an A to G substitution at nucleotide position 1639. The isoleucine at codon 547 is replaced by valine, an amino acid with highly similar properties, and is located in the cbEGF-like #4 domain. This amino acid position is not well conserved in available vertebrate species, and valine is the reference amino acid in other vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
