Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_000138.5(FBN1):c.2308G>A (p.Val770Ile), citing Ambry Variant Classification Scheme 2023: The p.V770I variant (also known as c.2308G>A), located in coding exon 19 of the FBN1 gene, results from a G to A substitution at nucleotide position 2308. The valine at codon 770 is replaced by isoleucine, an amino acid with highly similar properties, and is located in the cbEGF-like #08 domain. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
