Several computational tools predict a significant impact on normal splicing: Four predict the variant abolishes the canonical 5' splicing donor site. However, these predictions have yet to be confirmed by functional studies. Splice site mutations affecting canonical splice sequence or shown to alter splicing on mRNA/cDNA level are listed among criteria supporting a diagnosis of Marfan syndrome within the Ghent nosology (Loeys_2010). The variant was absent in 251120 control chromosomes. c.7330+1G>A has been reported in the literature in individuals affected with Marfan Syndrome (example, Attanasio_2008, Stheneur_2009). To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. One clinical diagnostic laboratory has submitted clinical-significance assessments for this variant to ClinVar after 2014 without evidence for independent evaluation and classified the variant as pathogenic. Based on the evidence outlined above, the variant was classified as pathogenic.
