Pathogenic for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_000138.5(FBN1):c.7330+1G>A, citing Ambry Variant Classification Scheme 2023. This variant lies in the FBN1 gene (transcript NM_000138.5) at the canonical splice donor site of the intron immediately after coding-DNA position 7330, where G is replaced by A; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: The c.7330+1G>A intronic pathogenic mutation results from a G to A substitution one nucleotide downstream of coding exon 58 of the FBN1 gene. This alteration has been detected in individuals with Marfan syndrome, and was reported to cause exon skipping by RNA studies (Attanasio M et al. Clin. Genet., 2008 Jul;74:39-46; Stheneur C et al. Eur. J. Hum. Genet., 2009 Sep;17:1121-8). In addition to the clinical data presented in the literature, alterations that disrupt the canonical splice site are expected to cause aberrant splicing, resulting in an abnormal protein or a transcript that is subject to nonsense-mediated mRNA decay. As such, this alteration is classified as a pathogenic mutation.
