NM_000393.5(COL5A2):c.247G>T (p.Ala83Ser) was classified as Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.A83S variant (also known as c.247G>T), located in coding exon 2 of the COL5A2 gene, results from a G to T substitution at nucleotide position 247. The alanine at codon 83 is replaced by serine, an amino acid with some similar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6503 samples (13006 alleles) with coverage at this position. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clincal significance of this variant remains unclear.

Protein context (NP_000384.2, residues 73-93): KIECQDVLDC[Ala83Ser]DPVTPPGECC
