Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_000393.5(COL5A2):c.3246G>C (p.Gln1082His), citing Ambry Variant Classification Scheme 2023: The p.Q1082H variant (also known as c.3246G>C), located in coding exon 46 of the COL5A2 gene, results from a G to C substitution at nucleotide position 3246. The glutamine at codon 1082 is replaced by histidine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
