Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_000093.5(COL5A1):c.803G>T (p.Gly268Val), citing Ambry Variant Classification Scheme 2023: The p.G268V variant (also known as c.803G>T), located in coding exon 6 of the COL5A1 gene, results from a G to T substitution at nucleotide position 803. The glycine at codon 268 is replaced by valine, an amino acid with dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
