Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_000093.5(COL5A1):c.5036C>A (p.Thr1679Lys), citing Ambry Variant Classification Scheme 2023: The p.T1679K variant (also known as c.5036C>A), located in coding exon 63 of the COL5A1 gene, results from a C to A substitution at nucleotide position 5036. The threonine at codon 1679 is replaced by lysine, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
