Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_000090.4(COL3A1):c.4394T>G (p.Phe1465Cys), citing Ambry Variant Classification Scheme 2023: The p.F1465C variant (also known as c.4394T>G), located in coding exon 51 of the COL3A1 gene, results from a T to G substitution at nucleotide position 4394. The phenylalanine at codon 1465 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
