Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_000090.4(COL3A1):c.3217G>A (p.Ala1073Thr), citing Ambry Variant Classification Scheme 2023: The p.A1073T variant (also known as c.3217G>A), located in coding exon 44 of the COL3A1 gene, results from a G to A substitution at nucleotide position 3217. The alanine at codon 1073 is replaced by threonine, an amino acid with similar properties. This amino acid position is poorly conserved in available vertebrate species, and threonine is the reference amino acid in other vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
