NM_004387.4(NKX2-5):c.85G>A (p.Ala29Thr) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.A29T variant (also known as c.85G>A), located in coding exon 1 of the NKX2-5 gene, results from a G to A substitution at nucleotide position 85. The alanine at codon 29 is replaced by threonine, an amino acid with similar properties. This amino acid position is poorly conserved in available vertebrate species, and threonine is the reference amino acid in other vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
