NM_001148.6(ANK2):c.3268A>G (p.Lys1090Glu) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.K1090E variant (also known as c.3268A>G), located in coding exon 29 of the ANK2 gene, results from an A to G substitution at nucleotide position 3268. The lysine at codon 1090 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
